Optimal temperature (enzyme)

The temperature at which enzyme activity peaks. Exceeding this causes thermal denaturation and rapid loss of activity.
Optimal temperature is the temperature at which an enzyme works most efficiently. Rising temperature increases molecular movement and reaction speed, but beyond a certain point, protein structure collapses (thermal denaturation) and function is irreversibly lost.
Koji amylase's optimal temperature is 50-60°C; protease is 40-50°C. Incubating amazake at around 55°C targets amylase's optimum. Enzymes denaturing during miso pasteurization occurs because heating far exceeds optimal temperatures.
Knowledge of optimal temperatures directly informs practical decisions about 'which temperature to choose' at each fermentation stage. Higher temperatures speed reactions but increase the risk of enzyme destruction—this trade-off is the essence of temperature management.
